ASSESSMENT OF THE OPTIMUM CONDITIONS FOR PRODUCTION AND PURIFICATION OF SUPEROXIDE DISMUTASE FROM ACTINOMYCETES

Abstract
Two actinomycetes, Streptomyces violaceoruber (S. violaceoruber) and Streptomyces spiroverticillatus (S. spiroverticillatus), were chosen from four Actinomycetes exhibited a high intracellular superoxide dismutase (SOD) productivity. The maximum productivity of SOD from both S. violaceoruber and S. spiroverticillatus was at 6 days of incubation, 35 °C and pH 7.0. The best SOD productivity from S. violaceoruber was at 0.05 M of K2HPO4 and from S. spiroverticillatus at 0.005 M. The maximum productivity of SOD from both Actinomycetes was achieved from media supplemented with lactose at 4- days of incubation period and to a lesser extent glucose, sucrose and maltose. Administration of trace elements to the culture media of S. violaceoruber and S. spiroverticillatus highly reduced the productivity of the enzyme. However, addition of FeCl2 and MnCl2 to the culture media that contain FeSO4 highly induced the best productivity of SOD at 8 days of incubation period compared to the control but still less than 6 days of incubation. The specific activity increased from 1585 to 8387 U/ mg protein and from 1539 to 10398 U/mg protein for S. violaceoruber and S. spiroverticillatus, respectively and the fold increased from 1.6 to 5.3 and from 1.6 to 6.7 for S. violaceoruber and S. spiroverticillatus, respectively.
